Background
This gene encodes a subunit of mitochondrial ATP synthase. Mitochondrial ATP synthase catalyzes ATP synthesis, using an electrochemical gradient of protons across the inner membrane during oxidative phosphorylation. ATP synthase is composed of two linked multi-subunit complexes: the soluble catalytic core, F1, and the membrane-spanning component, Fo, comprising the proton channel. The catalytic portion of mitochondrial ATP synthase consists of 5 different subunits (alpha, beta, gamma, delta, and epsilon) assembled with a stoichiometry of 3 alpha, 3 beta, and a single representative of the other 3. The proton channel consists of three main subunits (a, b, c). This gene encodes the alpha subunit of the catalytic core. Alternatively spliced transcript variants encoding the different isoforms have been identified. Pseudogenes of this gene are located on chromosomes 9, 2, and 16.

ATPA(529) was immunoprecipitated from 1mg of Jurkat cells membrane fraction, blotted with M25065 of 10μg. Western blot was performed from the immunopre-cipitate using M25065 at 1/1000 dilution. Anti-Mouse-IgG(HRP), specific to the non-reduced form of IgG, was used as secondary antibody at 1/10000 dilution. Lane 1: Jurkat cells membrane fraction. Lane2: IP product of Jurkat cells membrane fraction. Predicted band size : 60 KDa Observed band size : 60 KDa Blocking and dilution buffer and concentration:5% milk/TBST.

Flow cytometric analysis of 2% paraformaldehyde-fixed Jurkat (Human T cell leukemia cells from peripheral blood) cells labeling ATPA(529) with M25065 at 1/200 dilution (red) compared with a mouse monoclonal IgG isotype control (blue) and an unlabelled control (cells without incubation with primary antibody, green). Goat anti-mouse IgG (FITC) at 1/300 dilution was used as the secondary antibody.
